چکیده

Irrespective of the form of vitamin A fed to rats, it appears in blood almost exclusively as retinyl palmitate, and the liver always stores only this ester. Differences in the fatty acid composition of the dietary fats do not seem to have any influence on the type of retinyl ester absorbed and stored by the rat (Mahadevan & Ganguly, 1961; Mahadevan, Seshadri Sastry & Ganguly, 1963). It was therefore suggested that probably some specific lipoprotein transports retinyl palmitate from the intestine and stores it in the liver of rats (Mahadevan & Ganguly, 1961). If this were so, the intracardial injection of different retinyl esters into rats should lead to ready deposition in the liver of the palmitate in preference to the other esters. It is shown in the present paper that, when different retinyl esters are injected, the palmitate is more readily taken up by the liver, whereas, although small amounts of the other injected esters can be found in this tissue shortly after the injection, in the course of time they are all stored as the palmitate.

Hydrolysis of retinyl esters by non-specific carboxylesterases from rat liver endoplasmic reticulum.

The four most important non-specific carboxylesterases from rat liver were assayed for their ability to hydrolyse retinyl esters. Only the esterases with pI 6.2 and 6.4 (= esterase ES-4) are able to hydrolyse retinyl palmitate. Their specific activities strongly depend on the emulsifier used (maximum rate: 440 nmol of retinol liberated/h per mg of esterase). Beside retinyl palmitate, these este...

Hepatic uptake and metabolism of chylomicron retinyl esters: probable role of plasma membrane/endosomal retinyl ester hydrolases.

Previous studies have indicated the presence of both neutral and acid, bile salt-independent retinyl ester hydrolases associated with plasma membrane and endosome fractions of rat liver homogenates. In the present studies, chylomicrons containing tritium-labeled retinyl esters were injected intravenously into rats in order to study the initial metabolism of retinyl esters during and after uptak...

Tissue Distribution and Metabolism of Newly Absorbed Vitamin a in the Rat.

Chylomicrons containing newly absorbed labeled vitamin A were injected intravenously into normal intact rats, and the tissue distribution of radioactivity was observed for several days. Chylomicrons were used so that the vitamin could be administered physiologically, in the form in which it is normally absorbed. ‘The total recovery of lipid-soluble radioactivity in the entire animal varied from...

Neutral and acid retinyl ester hydrolases associated with rat liver microsomes: relationships to microsomal cholesteryl ester hydrolases.

We recently reported the presence of a neutral, bile salt-independent retinyl ester hydrolase (REH) activity in rat liver microsomes and showed that it was distinct from the previously studied bile salt-dependent REH and from nonspecific carboxylesterases (Harrison, E. H., and M. Z. Gad. 1989. J. Biol. Chem. 264: 17142-17147). We have now further characterized the hydrolysis of retinyl esters b...

Effects of Vitamin A in Neonates and Young Infants

Vitamin A is the generic name given to a group of fat-soluble compounds including retinol (the alcohol form), retinyl esters, retinaldehyde and retinol acid. Deficiency, first recognized in 1912, can damage the epithelial cells lining the respiratory tract. It can also affect immunocompetence, the reproductive function, growth and vision. The dose of vitamin A for neonates is 5,000 IU given int...
